CALCULATION OF PERCENTAGE OF VOLUME INFARCTED

FOR DOXY PIGS 04-A THROUGH 04-F

SINGLE RING SLICE WITH ALL BEADS IN SLICE

THREE PICTURES PER SLICE, APICAL SIDE, BASAL SIDE, AND THICKNESS OR SIDE VIEW

 

For apical and basal sides of slice:

  1. Use ImageJ (calibrated for x pixels/cm)
  2. Measure surface area of infarct (draw purple ring)
  3. Measure surface area of epi (draw green ring)
  4. Measure surface area of endo (draw yellow ring)
  5. Subtract surface area of endo from surface area of epi to get surface area of tissue
  6. Green area – yellow area = total surface area of tissue

  7. Measure thickness of slice with ImageJ taking a mean of five different measurements
  8. Average surface area of infarct of basal side and infarct of apical side
  9. Multiply mean surface area of infarct by thickness of ring to get volume of infarct
  10. Average total surface area of basal side total surface area of apical side
  11. Multiply average found in step 9 by the thickness of the slice to get the total volume of the tissue in the slice
  12. Divide the volume of the infarct (found in step 8) by the total volume of the tissue (found in step 10) to find the percentage of the volume infarcted
